Elution Buffers (TE, EB, Water)


Final Buffer Concentration: 

  • 10 mM Tris-Cl (pH 8.0) 

  • 1 mM EDTA (TE only)


Equipment & Consumables


Protocol:

For Mixing 200 ml of TE/EB Buffer:

  1. Dissolve 242.28mg Tris Base in 100mL Autoclaved dH20.

    • Note: Some EB recipes use 5 mM Tris-Cl, so you can choose to add instead 121.14g. I have not noticed a difference.

  2. Add 58.448mg EDTA (Only if you're making TE - do not add if you wish to make EB)

  3. Balance pH to 8.0 with diluted HCl

  4. Filter sterilise or autoclave. Add 15 mL to a falcon tube for immediate use and refrigerate the rest.

Water

  1. It is water. Sterilise it with an autoclave after either reverse osmosis or distillation.
