Elution Buffers (TE, EB, Water)

Final Buffer Concentration: 

• 10 mM Tris-Cl (pH 8.0) 

• 1 mM EDTA (TE only)

Equipment & Consumables

• PPE: Gloves, Lab Coat and Goggles (at least!)

• Sterile Workspace

• Sterile Micropipettes and Tips

• Pipetteboi and tips

• Weighing Scale (accurate to to 0.01g)

  • Weigh boats (or folded paper)

• Filters & Syringes OR Autoclave

• Optional: Heated/Stirring Platform

• pH Meter (recently calibrated)

• Graduated Beaker

• Measuring Cylinder

• 4°C Refrigerator

• ddH20 (MilliQ or RO)

• Tris Base

• Hydrochloric Acid (HCl)

• EDTA (Ethylenediaminetetraacetic acid)

Protocol:

For Mixing 200 ml of TE/EB Buffer:

1. Dissolve 242.28mg Tris Base in 100mL Autoclaved dH20.

   • Note: Some EB recipes use 5 mM Tris-Cl, so you can choose to add instead 121.14g. I have not noticed a difference.

2. Add 58.448mg EDTA (Only if you're making TE - do not add if you wish to make EB)

3. Balance pH to 8.0 with diluted HCl

4. Filter sterilise or autoclave. Add 15 mL to a falcon tube for immediate use and refrigerate the rest.

Water

1. It is water. Sterilise it with an autoclave after either reverse osmosis or distillation.
